*New* Disposable Culture System for High Throughput Screening Of Process Parameters and Production Cell Lines

Application As Process Optimization Tool

• Cultures start in bioreactor under production conditions
• On the day of interest, cells from the production run are transferred into 50ml tubes
• Process parameters are then screened in the shaken system under defined conditions for gas transfer
• Best parameters are then verified in a bioreactor

Ventilation

Ventilation of the culture vessel helps to avoid oxygen limitation and affects the CO₂ exchange. Since the latter makes the pH highly sensitive to ventilation (accumulation versus stripping of CO₂), a defined and adaptable ventilation is desirable.

• Defined ventilation
• No formation of foam at any speed
• Flexible working volume with one type of tube
• Allows fast medium exchange following centrifugation
• Fed batch approaches are possible
• Each tube is a completely independent vessel
• Rapid sampling, whole tube can be sacrificed

  Maintains cell suspension cultures on any incubating shaker while also allowing full control of CO2 and O2 exchange rate using selectable perforations on the cap. Internal CO2 is shown to be adequately maintained in the cultures for days even in non CO2 incubation conditions.

  The disposable 50ml Tubes are centrifugable to 5000Xg and feature easy to write on labels and one-hand removable caps to allow for easy, rapid sampling from many units.
